Melanotan 1 (afamelanotide) is a synthetic 13-amino-acid analog of α-melanocyte-stimulating hormone with high selectivity for the melanocortin-1 receptor (MC1R). In-vitro and preclinical studies characterize it as an MC1R agonist that elevates intracellular cAMP in melanocytes, upregulating tyrosinase activity and eumelanin biosynthesis. Its amino-acid substitutions confer increased receptor affinity and metabolic stability relative to native α-MSH.
