Overview
Kisspeptin-10 is the C-terminal decapeptide (Tyr-Asn-Trp-Asn-Ser-Phe-Gly-Leu-Arg-Phe) of the larger KISS1 gene product, from which the biologically active kisspeptins are proteolytically derived. It belongs to the RFamide family of neuropeptides, sharing the conserved C-terminal Arg-Phe-amide motif required for receptor engagement. The decapeptide retains the receptor-binding activity of the longer kisspeptin-54 parent while offering a shorter, synthetically accessible reference sequence. It is produced synthetically for laboratory research use.
Mechanism as studied
The research literature characterizes kisspeptin-10 as an agonist at KISS1R (GPR54), a Gq/11-coupled receptor expressed on hypothalamic GnRH-producing neurons. Receptor engagement is associated with phospholipase-C activation, inositol-trisphosphate signaling, and mobilization of intracellular calcium, endpoints described in cultured neuronal and heterologous receptor systems. Preclinical studies link this signaling to depolarization of GnRH neurons and modulation of pulsatile gonadotropin-releasing-hormone output. Downstream effects on luteinizing-hormone and follicle-stimulating-hormone secretion are characterized at the level of the hypothalamic-pituitary-gonadal axis.

Handling & storage

It is typically reconstituted with bacteriostatic water, with the diluent volume chosen to reach the working concentration a given assay requires (e.g. adding 1 mL to a 10 mg vial yields 10 mg/mL). Swirl gently to dissolve rather than shaking or vortexing, and aliquot the reconstituted solution to limit freeze-thaw cycles.
Store the sealed lyophilized vial at −20 °C protected from light, where it is stable long-term. Once reconstituted, keep refrigerated at 2–8 °C and avoid repeated freeze-thaw.
