Overview
PT-141 (bremelanotide) is a synthetic cyclic heptapeptide and a deamidated metabolite of melanotan 2, differing from its parent principally by a C-terminal free acid in place of the primary amide. It retains the constrained lactam-ring architecture of the parent compound. Functionally it is characterized principally as an agonist at the centrally expressed melanocortin MC3R and MC4R subtypes. It is produced synthetically as a reference compound for research into central melanocortin signaling.
Mechanism as studied
The research literature characterizes PT-141 as an agonist at the centrally expressed melanocortin receptors MC3R and MC4R, which couple through Gs protein to adenylyl cyclase and cyclic-AMP generation. MC4R activation within hypothalamic circuits is the principal focus of mechanistic study. Preclinical work associates this receptor engagement with modulation of downstream central dopaminergic signaling. Its receptor selectivity and cyclization distinguish it pharmacologically from its non-selective melanocortin parent compound.

Handling & storage
Supplied lyophilized for laboratory reconstitution, typically in bacteriostatic water to the concentration a given assay requires (for example, 2 mL added to a 10 mg vial yields 5 mg/mL). Swirl gently to dissolve; avoid shaking or vortexing the solution. Prepare in a clean field and aliquot to limit freeze-thaw cycles.
Store sealed lyophilized vials at −20 °C protected from light for long-term stability. Keep reconstituted material refrigerated at 2–8 °C and avoid repeated freeze-thaw.
Certificates of analysis
Independent and in-house certificates published for this compound. Each reports identity and HPLC purity for the tested lot.
Order PT-141 (Bremelanotide)
