Overview
GLP-1S corresponds to semaglutide, an acylated analog of human glucagon-like peptide-1 (GLP-1). A C-18 fatty-diacid moiety attached through a linker confers albumin binding and an extended plasma half-life relative to native GLP-1. It is studied as a long-acting, selective GLP-1 receptor agonist reference compound.
Mechanism as studied
The research literature attributes its activity to selective agonism at the GLP-1 receptor, a class-B G-protein-coupled receptor. In pancreatic beta-cell models, receptor activation is associated with glucose-dependent cyclic-AMP signaling and insulin secretion; in preclinical models the literature also describes suppression of glucagon release, slowed gastric emptying, and reduced food intake mediated through hypothalamic and brainstem circuits.
