Overview
ARA-290 (cibinetide) is a synthetic linear peptide of eleven amino acids whose sequence reproduces a segment of the helix-B domain on the outer, hydrophilic face of the erythropoietin (EPO) molecule. Because this face does not contact the classical homodimeric EPO receptor, the peptide is characterized as engaging tissue-protective signaling without the erythropoietic activity of the parent hormone. It is produced synthetically for laboratory research use and is studied as a reference compound for innate-repair-receptor biology.
Mechanism as studied
The research literature characterizes ARA-290 as a selective ligand of the innate repair receptor (IRR), a heteromeric complex proposed to assemble from the EPO receptor and the β-common receptor (CD131) shared with GM-CSF, IL-3, and IL-5. Engagement of this heteromer is associated in cell and preclinical models with anti-inflammatory and cytoprotective signaling — modulation of pro-inflammatory cytokine release and activation of cell-survival pathways in stressed cells — while leaving the homodimeric EPO-receptor pathway that drives red-cell production unengaged.
Research context
Contexts in which this compound appears in the in-vitro and preclinical research literature:
- Innate-repair-receptor binding and β-common-receptor (CD131) association studies.
- In-vitro anti-inflammatory endpoints: pro-inflammatory cytokine modulation in cultured immune cells.
- Neuronal and other stressed-cell culture models examining cytoprotective signaling.
- Comparative studies against erythropoietin distinguishing tissue-protective from erythropoietic activity.
- Preclinical models of neuropathic and inflammatory tissue injury reported in the literature.
Handling & storage
Supplied lyophilized for reconstitution in the laboratory, typically with bacteriostatic water to the working concentration required by the assay (diluent volume set by the target mg/mL). Add diluent slowly down the vial wall and swirl gently to dissolve rather than vortexing. Prepare in a clean field and aliquot the reconstituted peptide to minimize freeze–thaw exposure.
Store the sealed lyophilized vial at −20 °C protected from light, where it is stable long-term. Once reconstituted, keep refrigerated at 2–8 °C, protect from light, and avoid repeated freeze–thaw cycles.
Certificates of analysis
A certificate of analysis for the current lot is in preparation and will be published in the certificate library. Lot documentation is available on request for this compound.
